According to the podcasts with the writers and producers, Jacob is the "him" that people keep mentioning. For example, Mikhail (eye patch guy) said the man who brought him to the island is "a magnificent man" so Kate said something about Ben, but Mikhail said Ben is not "him." Also, Danny (the guy that Juliet shot) said at one point that Jack was "not even on Jacob's list." And then there was the brainwashing scene when the phrase flashed "God loves you as He loved Jacob."
There are probably biblical meanings involved as well, though the only thing I know is that one of Jacob's children in the Bible was Benjamin, so it's possible that "he"/Jacob is Ben's dad on the show.

if you played the Lost Experience and hunted around the database Persephone had information that there was a Jacob Vanderfield who was in the Board of Directors for the Hanso Foundation. He was sent to jail for taking bribes from an Chinese smugglers and got out early by his possible connections.

But more than likely Jacob is the head honcho who we haven't been introduced to yet but we probably will be this season. He is the illusive "him" that they reference.. "He" is a great man. "he" brought me to the island. "He" will kill me if I fail the mission..

Many of the blog sites think that Jacob is a character already known to us..hiding among the survivors. Perhaps Jack.. since Jack woke up in the jungle alone and then magically ran to the wreck..some feel it was suspicous from the first moment. Others think that Bernard may be Jacob. But both of these theories seem far fetched...but then again this is Lost.. so who knows.
